Brief Introduction

李惠琳,博士,中山大學教授,博導。201712月通過中山大學百人計劃進入太阳集团1088vip工作。長期從事生物質譜研究,圍繞完整蛋白的直接測序及修飾位點定位、蛋白-藥物作用機制及蛋白非共價複合物的結構解析等科學難題開發質Top-Down MS譜新方法,以實現對蛋白複合物結構及藥物作用機制的深度探索。其研究成果發表在Nature Chemistry, Analytical Chemistry, J. Am. Soc. Mass Spectrom.等國際主流化學、分析及質譜類雜志。

Research Direction

1)整合結構質譜技術(Integrative Structural Mass Spectrometry)的開發及其在藥物作用分子機制、靶向藥物篩選及蛋白複合物結構分析中的應用

  • Native top-down MS:直接建立蛋白/蛋白複合物的序列、結構和功能之間的關聯關系
  • 氫氘交換質譜(HDX-MS):獲取配體結合位點及蛋白結構動态變化等信息
  • 交聯質譜(cross-linking MS):開發新型交聯劑的開發,用以研究蛋白結構及蛋白-蛋白相互作用研究
  • 表面标記質譜(surface labeling MS):獲取蛋白結構及蛋白-蛋白相互作用信息

2)Middle-down/top-down蛋白質組學技術探索蛋白翻譯後修飾(PTMs)在生命過程中的調控機制:

  • Middle-down蛋白組學新技術的開發:酶切方法及組學流程的優化
  • 應用Middle-down蛋白組學技術探索PTMs在生命體中的調控機制
